When he woke up in the jungle right after 815 crashed and immediately went to work saving others

When he cut Ben's kidney and sacrificed his chance of escape to save Kate and Sawyer

When he finally unveiled his and Juliet's plan to "blow the others to hell"

When he beat the crap out of Ben after Ben ordered the "deaths" of Sayid, Jin and Bernard

When Locke held a gun to him and threatened to kill him, and he responded with, "Then do it, John"

When he told MIB "John Locke told me I needed to stay" and shoved him into the water

When, after the sub explosion, Kate told him they needed to kill MIB and he responded "I know."

When Jack and Locke's group met up in the finale and Jack tells MIB, "I'm gonna kill you."

When Jack catches up to MIB, gives his war cry, and engages MIB in a final, epic battle

-Shooting Locke IN THE FACE, nevermind that the gun wasn't loaded.
-"I didn't say I was gonna do it - I just wanted you to understand how you're gonna die."
And of course the (listed) "we're gonna blow 'em all to hell" and subsequent thrashing of Ben.
