I'm a Brit living in New Zealand. I've been told my writing has an English voice so I've had to take that into consideration in the setting of my novel's and the character's backgrounds. Here we work very much with British English, and for so far I've targeted English readers i.e. my second novel is set in London though two of the main characters are Kiwi's and in the novel I've made something of the different words used i.e. rucksack and backpack, flipflops and jandles. Yep like Irish words go figure.
I've downloaded Results not Typical though not read it yet. I read a couple of the lower star reviews - I find those more telling of the reader/book than the 5 star ones and noticed a comment on typos, and I wondered then if that related to English/American spellings. It didn't stop me buying the book as I made the assumption that was it.
It is a difficult question and I don't have an absolute answer. I tend to think go with the setting, so in Results Not Typical then probably it should have been American words/spellings, but if your next book is set in Ireland or England I'd go with English spelling.
