From the very first episode, When they spoke about Backgammon.
At the start of season 2 as hinted by Desmond's "See you in another life brother".
At the end of season 2, When the hatch timer showed glyphs that translated to "Afterlife/Underworld"
Near the end of season 3, when they finally got an end date for the show.
They only thought of it whilst preparing season 6.
Near the end of season 6, When they couldnt figure out how to converge the 2 alternate realities.
