If the GOP vehemently oppose and fight Obama on his immigration plan, will that hurt them with Latino voters in upcoming elections?

Yes, they will look as if they oppose any immigration reform and it'll come back to bite them in 2016 and beyond when Latinos vote for the Dems.

No, even Latinos will regard Republicans as defending the US Constitution by trying to stop a president from excessively using executive orders.

I don't think it'll have a huge impact on how the Latino community will ultimately vote or turn out in future elections.
