I suppose, humans in question IS post.
If so, then measuring them in pure-human setting - pointless, mainly because their reality pattern include more conceptions, then whole humanity can imagine.
To the point - this makes all calculations in post №1 useless and senseless.
More to the point - conquering Galaxy (in any meanig af this word) IS something pure-humans will do (assuming they found a way - see calculations in post №1), not post-humans. Transcending through Prigozhin's (phase) barrier will change psychology and motivations of >H.
Call me optimistic, but I think, that transcends will use Galaxy primarily for studying and enjoyment, not colonization. They simply lose need for spread (in such crude way, if any).

    Some thoughts
1 radiation probably kills before leaving Solar system
2 a heavy space ship (pay load of > 1000 tonnes) cannot be launched in one
3 to build a ship in orbit (geostationary preferred) will be too expensive for any nation
4 Building and construction technology in space e.g. production of a carbon fibre beehive, is not present
5 building speed to 1 % of SoL requires much more boost energy than present rickets have. (
6 even nuclear power will have early limits (power reactors of nuclear aircraft carriers last a few decennia)
7 space ship needs at least 300+ people. may be a tenfold is required to represent and maintain all disciplines of science and skills
8 A biosphere to produce food for e.g 1000 years will be gigantical
9 there must be even more energy needed to brake the space vehicle when approaching a suitable solar system.
10 All tools/materials for humanification of a more or less suitable planet/moon must be on board
11 If a suitable planet is found within the habitable zone it might have life. We might not withstand their weapons or diseases.
12 seeds should be sent to all directions to improve the chances of success; the total amount of energy needed for 1000 missions exceeds the available stocks on Earth

The best is reducing the amount of people on Earth to such a number that hunanity is in ballance with nature.

With a stringent 1 child per couple regime (UN has to force this) it wil take some 150 years to have the same amount of people as in the times of the Roman Empire
